Our Core Prep service is useful for both new and seasoned researchers who would like to fast track their Sanger sequencing. This is an excellent option for researchers who are new to Sanger sequencing as our team are experienced in the process and will be able to assist you in your research. Our sequencing guide will provide you with an overview of the sequencing process and our services.

  • Core Prep –You submit a mix of purified DNA and primer. We perform the sequencing reaction and capillary separation. Turnaround time: 3-5 days.
  • Core Prep Plus –You submit unpurified PCR product and primer. We perform the PCR clean up, sequencing reaction and capillary separation. Turnaround time: 3-5 days.
  • Data analysis service –please contact us for further information.

Sample Submission Procedure

Please follow the sample submission guidelines carefully to ensure smooth processing and a quick turnaround time.

  • Create account/Login and submit samples online using the dnaLIMS system (refer to our LIMS User Guide).
  • Prepare a mixture of purified template and primer per sample for submission. The final ratio may require optimisation. Please refer to the table below as a starting point. Please note that the concentration listed in this table is the final concentration in the tube (i.e. 10ul).

  • Forward and reverse are considered as two individual samples and must be prepared separately.
  • PCR products must be purified prior to sequencing in order to remove residues from PCR. An additional ExoSAP-IT clean-up step is available via the Core Prep Plus option. Your samples may require other methods of purification, please see our guide to Sanger sequencing for more information.
  • For the Core Prep service (no clean up) template and primer for each sample should be combined in one tube or well, minimum volume of 10ul. Please send samples in 1.5ml tubes or AB3730 compatible plates.
  • For Core Prep Plus (Exosap-IT included) it is essential to send primers separately in 1.5mL tubes. Please send samples in 1.5ml tubes or AB3730 compatible plates.
  • It is important to select the correct “Service Request” option in the drop down menu, please select the Plate96- or Plate48- for plate submissions.  For Core Prep Plus submissions add the extra step of selecting “YES” to “Exosap-IT” during online submission. See image below.

  • Sample numbers for Core Prep or Core Prep Plus plate submissions are 40-47 for half plate (in odd columns) and 80-95 for full plate. A control sample will be included in each run. Please leave the last well (11H or 12H) empty for the control sample.
  • For individual samples, label tubes with the order number and consecutive numbering on the lid. For example, for order number 1234 with 3 samples, label tubes as “1234-1”, “1234-2” and “1234-3” (see below). Label plates with the order number or plate number.

  • Send samples by either courier or Express Post (Australia Post) to ensure prompt delivery. Please send samples and a copy of your submission confirmation to the address below. Alternatively, you can drop off your samples at our sample reception weekdays between 8am and 6pm.
  • You will receive an email when your results are available. You can access your results via dnaLIMS as per the LIMS user guide.

Data Delivery:

Once sequencing is complete you will receive an email notification. You can access your data via an online account. The following files are available to download:

  • Raw trace (.ab1 file)
  • Raw, unfiltered basecall sequence (.seq file)
  • Text file of you sequence
  • FASTA Sequence
  • System QC score of the sequence

Quality Guarantee:

  • We include internal controls on all sequencing runs to ensure quality data
  • Batches of Core Prep include a pGem and M13 primer control reaction
  • All sequencing runs include a sequencing standard control
  • Free re-run service if results do not meet our quality standards

Note: Working days are Monday – Friday. Samples must be received by 12 noon to be included in the days processing list.

 

  • Services Offered
